How about removable buttons, then you can have both, though you would have to add shanks to the antler for that.
But if you will only wear it alone or with a tweed jacket, then whatever buttons are on the tweed would be my suggestion.

I'm a fan of removable buttons. It shouldn't be too difficult to sew on a small ring to the back of the antler buttons in the same way as sewing them to the fabric. Though, a loop of fabric to go through the eyelet and a small safety pin to hold it might work just as well.
I tried to ask my inner curmudgeon before posting, but he sprayed me with the garden hose…

My instinct would be to go with black. I have a few waistcoats, and even though my favorite ones have horn and wood buttons (actually, my favorite one has leather buttons, but that's neither here nor there), they're mostly tweedy, themselves. A dressier one, a navy pinstripe, looks good with its black buttons; I associate the brighter (relatively) dyes of modern tartans and the navy with black, and the more neutral browns, &c., with antler. Along similar lines, antler buttons might not be contrastive enough, or, looking at it another way, might be too contrastive (depending on the color of the antler and the look you're going for).
Just my thoughts.
